目的:探讨下调mir-130a-3p的表达量后对RAB5B基因在耐药株MCF-7/ADR细胞中的影响。方法:采用CCK8检测法检测耐药细胞株MCF-7/ADR和MCF-7细胞的耐药性及存活率;采用qRT-PCR的方法验证MCF-7和MCF-7/ADR中mir-130a-3p、RAB5B的本底表达;采用转染mimic的方法下调mir-130a-3p基因验证其对在MCF-7和MCF-7/ADR细胞中对RAB5B的影响。结果:MCF-7/ADR细胞株耐药性和存活率好。MCF-7和MCF-7/ADR中均有mir-130a-3p和RAB5B的表达,mir-130a-3p在耐药株和非耐药株表达差异有统计学意义(p<0.05),但RAB5B在两株细胞中表达差异并无统计学意义(p>0.05),说明RAB5B基因对细胞的耐药性没有影响。结论:。mir-130a-3p在耐药株和非耐药株中表达有差异,但其靶基因RAB5B对细胞的耐药性并没有影响
